Isolation, proliferation, characterization and in vivo osteogenic potential of bone-marrow derived mesenchymal stem cells (rBMSC) in rabbit model.
Information on isolation, characterization of rabbit MSC and its evaluation in critical bone defect (CSD) is scarcely available. Here, we attempted to isolate, proliferate, differentiate, characterize and evaluate the in vivo osteogenic potential of bone marrow derived mesenchymal stem cells (BMSCs) collected from New Zealand White rabbits. They were isolated and proliferated in antibiotic supplemented DMEM (Dulbecco’s Modified Eagle’s media). ⋯ The rBMSCs were positive for CD44, CD73 and CD105 and negative for CD34 and CD45 and could differentiate to osteogenic cells in osteogenic induction media. The in vivo experiments in rabbit CSD model confirmed that rBMSCs promote faster healing of critical size defects. Hence, we may suggest that rBMSCs are suitable for bone formation in fracture healing and non-union.

Chronic spinal infusion of loperamide alleviates postsurgical pain in rats.
Plantar incision in rat generates spontaneous pain behaviour. The opioid drug, morphine used to treat postsurgical pain produces tolerance after long-term administration. Loperamide, a potent mu-opioid agonist, has documented analgesic action in various pain conditions. ⋯ However, the highest dose of morphine produced inhibition of thermal hyperalgesia till 5th day and mechanical allodynia only till 3rd day post-plantar incision. Morphine and loperamide produced analgesia in postsurgical pain, which may be mediated through different mechanisms. Longer duration of analgesia with loperamide could probably be due sustained blockade of calcium channels.

Intranasal administration of insulin lowers amyloid-beta levels in rat model of diabetes.
Alzheimer's disease (AD) is a neurodegenerative disorder characterized by abnormal accumulation of amyloid beta (A beta) peptide in brain regions subserving memory and other cognitive functions. Hyperglycemia and perturbed insulin signaling have been proposed as pathogenic factors contributing to AD. ⋯ Using this model, the effect of intranasal administration of insulin on reduction of A beta levels was measured. The current findings strengthen the case for insulin as therapy for AD afflicted individuals with or without diabetes.

Effect of nitric oxide in protective effect of melatonin against chronic constriction sciatic nerve injury induced neuropathic pain in rats.
Developing a successful treatment strategy for neuropathic pain has remained a challenge among researcher and clinicians. Various animal models have been employed to understand the pathogenic mechanism of neuropathic pain in experimental animals. The present study was designed to explore the possible nitric oxide mechanism in the protective effect of melatonin against chronic constriction injury (CCI) of sciatic nerve in rats. ⋯ Further, L-NAME (5 mg/kg) pretreatment with sub-effective dose of melatonin (2.5 mg/kg, ip) significantly potentiated melatonin's protective effect which was significant as compared to their individual effect per se. However, L-arginine (100 mg/kg) pretreatment with melatonin (2.5 mg/kg, ip) significantly reversed its protective effects. Results of the present study suggest the involvement of nitric oxide pathway in the protective effect of melatonin against CCI-induced behavioral and biochemical alterations in rats.

Schwann cells promote neurite outgrowth of dorsal root ganglion neurons through secretion of nerve growth factor.
The transplantation of Schwann cells (SCs) could successfully promote axonal regeneration. This is likely to attribute to the adhesion molecules expression and growth factors secretion of SCs. But which factor(s) play a key role has not been precisely studied. ⋯ The high concentration of NGF protein was detected in SC-CM. When K-252a, a specific inhibitor of NGF receptor was added, DRG neurite outgrowth was significantly decreased in a concentration-dependent manner. These data strongly suggest that SCs play important roles in neurite outgrowth of DRG neurons by secreted NGF.
