Sera from 3 rabbits and 1 goat which contained precipitating antibodies to highly purified rat CNS myelin proteolipid protein (PLP) did not inhibit CNS myelin formation nor destroy intact CNS myelin in cultures of rat spinal cord. CNS myelin which formed in the presence of antiPLP serum and complement appears ultrastructurally identical to myelin formed in normal control cultures. The lack of demyelinating and myelin formation inhibiting activities in antisera to PLP suggests either: (1) PLP is not exposed on the external surface of the myelin membrane; or (2) the externally exposed portions of PLP were rendered nonantigenic during the purification of PLP.

Using biochemical parameters the present study sought to assess the normal developmental pattern of the dopaminergic innervation of the olfactory tubercle (OT) and how it is affected by olfactory bulbectomy. In rats, the adult pattern of cellular organization is achieved in the OT gradually over the first 7 days after birth. On the other hand, tyrosine hydroxylase (TH) and [3H]dopamine ([3H]DA) uptake, while present at low levels, start to increase rapidly only after the first 7 days reaching adult levels by 40 and 20 days after birth, respectively. ⋯ However, bulbectomy in 1-day-old rats did not produce any significant changes 40 days later. The findings suggest that during postnatal growth TH activity is increased in DA cell bodies, preceding the changes in DA terminals of the OT, resembling the changes occurring during collateral sprouting in adults. In addition, changes indicative of collateral sprouting do not occur in response to deafferentation of the OT in 1-day-olds but do in 10-day-olds or older animals, a phenomenon probably related to a critical development period of the OT.

In cerebellum, excitatory and inhibitory responses of Purkinje cells, produced both synaptically and by microiontophoresis of putative amino acid neurotransmitters, have been shown previously to be enhanced during NE iontophoresis. The influence of locus coeruleus conditioning stimulation on Purkinje cell responses to GABA iontophoresis was examined to determine whether endogenous NE, released from synaptic terminals, could exert similar modulatory effects. ⋯ Repetitive activation of the classic non-adrenergic cerebellar afferents did not enhance the GABA response, despite causing a direct depression in spontaneous rate. A neuromodulatory role is suggested for tonic adrenergic input in the mammalian central nervous system.

The sensation of pain and the nociceptive flexion reflex of the biceps femoris muscle (RIII, Bi) elicited by electrical stimulation of the ipsilateral sural nerve were studied in human during 4 conditions: (1) a mental task; (2) a stress; (3) during noxious stimulation of the contralateral ulnar nerve; (4) after an intense noxious stimulation of the sural nerve itself. An inhibition of both pain sensation and RIII, Bi was observed in (1), while a facilitation of these parameters was noted in (2). In contrast, no change in RIII, Bi occurred in (3) while the pain sensation was inhibited. ⋯ The latter recovered to its control values 28--30 sec later. These results show the possibility of a dissociation between afferent ascending nociceptive messages and nociceptive motor activity (in 3 and 4). They suggest that supraspinal descending influences can act differently on spinal dorsal horn neurons in the case of pain ascending volleys, and in the case of spinal nociceptive motor activity.
