Hyperinsulinemia and impaired glucose tolerance are associated with liver cirrhosis. To investigate whether insulin-degrading activity in liver tissue plays a role in hyperinsulinemia, we assayed this activity in biopsy tissue from healthy and cirrhotic subjects. ⋯ Therefore, insulin-degrading activity does not appear to be involved in the hyperinsulinemia that occurs in liver cirrhosis. The study provides indirect evidence that hyperinsulinemia and impaired glucose metabolism in liver cirrhosis are due to different mechanisms (receptorial and post-receptorial defects, and altered feedback inhibition of insulin secretion).

The radio enzyme assay for histamine based on the transmethylation with purified histamine-N-methyltransferase and utilizing [3H-methyl]-S-adenosylmethionine as the methyl donor has been optimized to measure low histamine concentrations, for example in plasma. The pH-optimum for the assay is pH 8.3 in Tris-glycine buffer at 20 degrees C. ⋯ EDTA and dithiothreitol were included in the assay to keep the histamine-N-methyltransferase active as agents that oxidize -SH groups were found to be inhibitory to the reaction. The present assay is sensitive to about 0.5 nmol/l of histamine in a sample volume of 50 microliter (about 3 pg/sample).

Diode-array spectrophotometry for simultaneous measurement of hemoglobin pigments.
A prototype oxygen saturation meter was used to measure the concentrations of deoxygenated hemoglobin (rHb), oxyhemoglobin (HbO2), carboxyhemoglobin (HbCO), methemoglobin (MetHb), and sulfhemoglobin (SHb) in 35 microliter blood. Simultaneous absorbance measurements at 535, 560, 577, 622, 636, and 670 nm permitted the composition of any hemoglobin pigment mixture to be determined more accurately, precisely and easily than before. The inclusion of 670 nm, where the hemoglobin pigments have low absorption coefficients, allowed correction for turbidity.

In order to study the relationship between sex, age and glucose, and the concentrations of various fuel related blood substrates in children during prolonged fasting, we have selected data of fasting procedures in 13 control children aged 3-5 yr, fasted 24 h, and 58 control children aged 6-15 yr, fasted 40 h. Compared to the blood results after overnight fast, glucose is decreased, and lactate, pyruvate, ketones and non-esterified fatty acids (NEFA's) are all clearly increased at the end of fast. The concentrations of alanine and triglycerides remain unchanged. ⋯ Ketones are negatively related with glucose, whereas pyruvate, NEFA's and triglycerides are not glucose-related. Lactate and alanine are weakly related to glucose. The data demonstrate diminished glucose homeostasis and increased ketogenesis in younger children compared to older ones during prolonged fasting.

The correlation between human plasma melatonin levels and urinary 6-hydroxymelatonin excretion.
A significant correlation (0.76) has been found between nighttime peak plasma melatonin levels and the 24-h urinary excretion totals for conjugated 6-hydroxymelatonin for a group of 22 women. This study validates the comparison of plasma levels of the hormone or urinary levels of its metabolite to assess pineal gland production of melatonin in humans.
