Many studies have investigated test-retest reliability of active voxel classification for fMRI, which is increasingly important for emerging clinical applications. The implicit impact of voxel-wise thresholding on this type of reliability has previously been under-appreciated. This has had two detrimental effects: (1) reliability studies use different fixed thresholds, making comparison of results is challenging; (2) typical studies do not assess reliability at the individual level, which could provide information for selecting activation thresholds. ⋯ This variation depended strongly on the individual tested. Finally, we demonstrated novel procedures using ROC-r and overlap analysis to optimize thresholds on a case-by-case basis. Ultimately, a method to determine robust individual-level activation maps represents a critical advance for fMRI as a diagnostic tool.

Real-time segmentation of burst suppression patterns in critical care EEG monitoring.
Develop a real-time algorithm to automatically discriminate suppressions from non-suppressions (bursts) in electroencephalograms of critically ill adult patients. ⋯ Automated segmentation of burst suppression EEG patterns is an essential component of quantitative brain activity monitoring in critically ill and anesthetized adults. The segmentations produced by our algorithm provide a basis for accurate tracking of suppression depth.

Prolonged sleep fragmentation of mice exacerbates febrile responses to lipopolysaccharide.
Sleep disruption is a frequent occurrence in modern society. Whereas many studies have focused on the consequences of total sleep deprivation, few have investigated the condition of sleep disruption. ⋯ This study demonstrates that prolonged sleep disruption of mice exacerbates febrile responses to lipopolysaccharide. This device provides a method to determine mechanisms by which chronic insufficient sleep contributes to the etiology of many pathologies, particularly those with an inflammatory component.

Increased anesthesia time using 2,2,2-tribromoethanol-chloral hydrate with low impact on mouse psychoacoustics.
To examine psychoacoustics in mice, we have used 2,2,2-tribromoethanol anesthesia in multiple studies. We find this drug is fast-acting and yields consistent results, providing 25-30 min of anesthesia. Our recent studies in binaural hearing prompted development of a regimen to anesthesia time to 1h. We tested a novel cocktail using 2,2,2-tribromoethanol coupled with low dose chloral hydrate to extend the effective anesthesia time. ⋯ We demonstrate that 375 mg/kg 2,2,2-tribromoethanol followed after 5 min by 200mg/kg chloral hydrate provides an anesthesia time of 60 min, has negligible effects on ABR wave latencies and thresholds and non-systematic effects on amplitudes.

A modified Hargreaves' method for assessing threshold temperatures for heat nociception.
This study describes a modified Hargreaves' method for assessing paw withdrawal threshold temperatures for heat (PWT-H) nociception in the hind paws of rats. This method utilises radiant heat to maintain controlled lamp temperatures (CLTs) on a glass floor beneath the rat hind paw. An ascending series of CLTs were applied for 10s, with 5-10min intervals between applications, until characteristic withdrawal behaviour was observed or a cutoff CLT was reached. ⋯ Three hours following plantar incision, the PWT-H decreased to 37.5±0.2°C, consistent with previous observations of C-fibre afferents from incised glabrous skin firing at 36.7±3.6°C. Parallel testing, using the current method and an electronic von Frey device, illustrated similar degrees of incision-induced hyperalgesia, gradual improvements in hyperalgesia, and reversals induced through morphine and gabapentin. In conclusion, the present method facilitates a comparison of PWT-H using electrophysiological and human psychophysical studies involving thermosensation, and as a behavioural assay identical to von Frey testing, this method also measures the threshold for nociception.
