Measuring tidal volume (VT) in nonintubated swine or swine with leaking breathing circuits is challenging. The aim of this study was to validate respiratory inductance plethysmography (RIP) for measuring VT in swine that are comparable in size to adult humans. To determine calibration curves, VT and RIP readings were obtained from anesthetized swine (n = 8; weight, 46-50 kg) during positive-pressure (mechanical) ventilation and spontaneous breathing. ⋯ The data revealed that VT was linearly correlated with the movements of both ribcage and abdomen as measured by using plethysmography over a large range of tidal volume (44 to 1065 mL). In addition, the intercept of the linear equation was small or even negative during spontaneous breathing but increased significantly (maximum, 145 mL, 59.2 ± 35.1 mL) during positive pressure ventilation. Our results indicate that VT in swine can be calculated by using a simple univariate linear regression equation with RIP readings obtained during either mechanical ventilation or spontaneous breathing.

Effects of buprenorphine and estrous cycle in a murine model of cecal ligation and puncture.
The effect of opioids on the immunopathology of sepsis models in mice has been controversial. In previous work, we showed that mortality and various inflammatory parameters did not differ between female mice given saline or buprenorphine after cecal ligation and puncture. To investigate further, we hypothesized that buprenorphine would not affect outcomes of sepsis at any stage of estrous. ⋯ At 24 h, buprenorphine-treated mice in proestrus had more circulating neutrophils and monocytes than did saline-treated mice in proestrus and more circulating WBC than did mice in any other stage with or without buprenorphine. Our current results suggest that the effects of buprenorphine on a 50% survival model of sepsis in BALB/c female mice are minimal overall but that the stage of estrous has various effects in this model. Investigators should consider the effects of buprenorphine and estrous cycle when using female mice in sepsis research.

The purpose of this work was to develop and characterize an aortopulmonary shunt model of chronic pulmonary hypertension in swine and provide sequential hemodynamic, angiographic, and histologic data by using an experimental endoarterial biopsy catheter. Nine Yucatan female microswine (Sus scrofa domestica) underwent surgical anastomosis of the left pulmonary artery to the descending aorta. Sequential hemodynamic, angiographic, and pulmonary vascular samples were obtained. ⋯ Autopsy confirmed severe pulmonary vascular changes, including concentric obstructive neointimal and plexiform-like lesions. This swine model showed hemodynamic, angiographic, and histologic characteristics of chronic pulmonary arterial hypertension that mimicked the arterial pulmonary hypertension of systemic-to-pulmonary arterial shunts in humans. Experimental data obtained using this and other models and application of an in vivo endoarterial biopsy technique may aid in understanding mechanisms and developing therapies for experimental and human pulmonary arterial hypertension.
