Review[Progress on role of extracellular ATP and its metabolite adenosine in immunoregulation: Review].
Extracellular adenosine triphosphate (ATP), as an extracellular messenger, participates in the immune response and inflammatory process, and is considered as a dangerous signal molecule. On one hand, extracellular ATP promotes inflammation through activating ATP receptor represented by P2X7 (P2 purinergic receptor) and downstream NLRP3 inflammasome assembly. On the other hand, it plays an anti-inflammatory role through conversion to adenosine by CD39 and CD73 on the cell surface and acting via adenosine receptor (P1 purinergic receptor). ⋯ Recent studies have shown that as endogenous tissue-derived signal molecules, extracellular ATP and its metabolite adenosine play a vital role in immunoregulation through purinergic metabolic pathway. The change of ATP and adenosine concentration in tissue microenvironment can affect the occurrence and resolution of inflammation, which has guiding significance for exploring the prevention and treatment strategies of inflammatory diseases. In this review, we summarize that CD39/CD73 synergistically regulates the balance of extracellular ATP and adenosine, thus influencing immune cell functions through P2 receptor and P1 receptor signaling pathway.

[Exploration and practice of online teaching system of medical immunology based on small private online course (SPOC) model during coronavirus disease 2019 (COVID-19) pandemic].
During the pandemic of coronavirus disease 2019 (COVID-19), the Chinese Ministry of Education put forward the requirement of turning offline education into online teaching to universities. Our department positively responded and rapidly mobilized to establish an online teaching system of medical immunology based on the small private online course (SPOC) model, which has a main body of the SPOC model with "video course as the main part, supplemented by online Q&A". ⋯ In this process, our department continuously summarizes experiences and discovers problems. Herein, we generalize and sort out the exploration and practice of constructing the online teaching system, to carry out offline teaching after the pandemic and to continue this teaching model in the future, for reference and guidance.

[Tanshinone IIA attenuates hydrogen peroxide-induced senescence of human umbilical vein endothelial cells through activating SIRT1/eNOS pathway].
Objective To explore the effect of tanshinone IIA (TSA) on hydrogen peroxide (H2O2)-induced senescence of human umbilical vein endothelial cells (HUVECs) and the underlying mechanism. Methods HUVECs were cultured in vitro and divided into the control group, model group and TSA group. The cells in the TSA group were pre-treated with TSA for 24 hours. ⋯ Conversely, the expression of P21 and P16 proteins in the TSA group were lower than those in the model group, and SIRT1 and p-eNOS/eNOS were higher in the TSA group than those in the model group. Transfected with SIRT1 siRNA significantly down-regulated the expression of SIRT1 in HUVECs and the positive rate of SA-β-gal staining was notably raised when SIRT1 was silenced in TSA-treated HUVECs. Conclusion TSA attenuates H2O2-induced endothelial cell senescence by activating SIRT1/eNOS signaling pathway.

[Estradiol increases the level of myocardial voltage-gated calcium channel α1C subunit (CACNA1C) in septic mice].
Objective To investigate the effect of estradiol on myocardial voltage-gated calcium channel α1C subunit (CACNA1C) in septic mice. Methods Estradiol benzoate was injected subcutaneously into male mice followed by sepsis induced by cecal ligation and puncture (CLP). The experimental mice were divided into 4 groups: sepsis group, estradiol-treated sepsis group, sham group, estradiol-treated sham group. 3, 6, 12, 24 hours after CLP, the mRNA level of CACNA1C in left ventricular was detected by real-time fluorescent quantitative PCR, the protein expression level of CACNA1C was detected by Western blot analysis. ⋯ Results The expression of CACNA1C was significantly decreased in sepsis group at 6 hours and remained at this low level at 12, 24 hours. The expression of CACNA1C in estradiol-treated sepsis group did not change at 3, 6 and 12 hours, but was significantly decreased at 24 hours. Conclusion Exogenous estradiol can increase the left ventricular level of CACNA1C in septic mice.

[Establishment of Acinetobacter baumannii-induced pneumonia model in mice].
Objective To establish Acinetobacter baumannii (A. baumannii)-induced pneumonia models in C57BL/6 mice, and study the molecule mechanism of A. baumannii infection. Methods Eighty C57BL/6 mice were divided into normal control group, cyclophosphamide-treated group, A. baumannii infection group, and cyclophosphamide-pretreated A. baumannii infection group. Immunodeficient mice were prepared by injecting cyclophosphamide intraperitoneally. ⋯ The cytokines of blood increased markedly 6 hours post infection, and then decreased until 72 hours. However, the cytokines continuously increased in cyclophosphamide-pretreated A. baumannii infection group. Conclusion A. baumannii-induced pneumonia models in C57BL/6 mice were established successfully.
