Enhanced TLR4 Expression on Colon Cancer Cells After Chemotherapy Promotes Cell Survival and Epithelial-Mesenchymal Transition Through Phosphorylation of GSK3β.
Phosphorylation of glycogen synthase kinase 3β (GSK3β) by phosphatidyl-inositide 3-kinase (PI3K)/protein kinase B (AKT) or inhibition of GSK3β with small-molecule inhibitor attenuates cell survival and proliferation and increases apoptosis in most cancer cell lines. In this study, we investigated the role of phosphorylated GSK3β activated by enhanced toll-like receptor 4 (TLR4) expression in drug-treated colon cancer cells as a model of post-chemotherapy cancer cells. ⋯ These results suggest that TLR4-induced GSK3β and ERK phosphorylation independently controls cancer cell survival and regulation of GSK3β and ERK after chemotherapy, making TLR4 a critical target for reducing drug resistance and metastasis in patients with colon cancer.

Ifosfamide and Etoposide Chemotherapy in the Treatment of Recurrent/Refractory Rhabdomyosarcoma in Adults.
No standard chemotherapy for adults with recurrent/refractory rhabdomyosarcoma (RMS) has yet been established. The present study aimed to assess the effect of ifosfamide and etoposide (IE) chemotherapy on previously treated RMS. ⋯ IE chemotherapy could be an alternative optional treatment method in adults with recurrent/refractory RMS.

Third-line Enzalutamide Following Docetaxel and Abiraterone in Metastatic Castrate-resistant Prostate Cancer.
There are no published randomised trials on the efficacy of enzalutamide against metastatic castrate-resistant prostate cancer (mCRPC) after docetaxel and abiraterone. We evaluated the activity of third-line enzalutamide in men with mCRPC after docetaxel and abiraterone. ⋯ In mCRPC, enzalutamide has modest activity after docetaxel and abiraterone. Response to previous abiraterone is not predictive of subsequent enzalutamide response.

Preoperative Chemotherapy with Bevacizumab Extends Disease-free Survival After Resection of Liver Metastases from Colorectal Cancer.
The benefit of preoperative chemotherapy for patients with liver metastases from colorectal cancer remains unclear. We evaluated the efficacy of preoperative chemotherapy with bevacizumab in such patients, and attempted to identify clinical predictors of recurrence. ⋯ Preoperative chemotherapy with bevacizumab appears to be an effective treatment modality for liver metastases from colorectal cancer, and results in prolonged DFS.

Efficacy and Safety of TAS-102 in Clinical Practice of Salvage Chemotherapy for Metastatic Colorectal Cancer.
TAS-102 is an anti-metabolite which demonstrated activity against multidrug-resistant advanced colorectal cancer. Its major toxicities are hematological disorders. ⋯ TAS-102 was safely administered to modestly fragile patients with equivalent efficacy to that for the non-fragile population. Further investigation of sequential treatment using regorafenib and TAS-102 is needed.
