The combined use of thermal desorption and selected ion flow tube mass spectrometry for the quantification of xylene and toluene in air.
Thermal desorption (TD) is commonly employed for volatile chemical analysis, it being the method of choice for occupational health and safety monitoring. TD allows for offline capture of volatiles onto a solid sorbent followed by desorption and analysis at a later time. Although TD is routinely used in conjunction with gas chromatography (TD-GC), the assay throughput is low and requires the use of gas standards for quantification. ⋯ Due to the robustness of the SIFT-MS analysis in the presence of water vapour and other major components of air, the purging of tubes usually required to remove these constituents during the TD cycle was not required, therefore reducing the TD cycle time. Comparing the quantity of xylene and toluene applied to the TD tube with the absolute levels quantified by SIFT-MS subsequent to desorption suggested a recovery of over 95% of the applied compound. We conclude that the combination of TD and SIFT-MS allows more rapid and accurate quantification of xylene and toluene (compared with TD-GC) to be achieved without the need for calibration standards, features which may be advantageous in applications requiring rapid analysis and high throughput.

A strategy for high-throughput analysis of levosimendan and its metabolites in human plasma samples using sequential negative and positive ionization liquid chromatography/tandem mass spectrometric detection.
Levosimendan (Simdax) is an approved drug in approximately 40 countries and currently in phase III clinical studies in the USA and Europe. An accurate, high-throughput and rugged assay is critical to support these clinical trials. Due to the mechanism of drug metabolism, the drug and its active metabolites often have significant differences in their chemical properties. ⋯ The inter-assay %CVs for QC samples were from 1.2% to 1.8% for levosimendan, 1.3% to 2.7% for OR-1855, and 1.4% to 3.4% for OR-1896. The mean % biases for QC samples were from 1.5% to 5.5% for levosimendan, -1.4% to 2.6% for OR-1855, and -0.3% to 4.5% for OR-1896. By using a single extraction approach coupled with sequential LC/MS/MS analysis for levosimendan and its metabolites, the assay maintained high throughput and low sample volume usage.

Determination of four tobacco-specific nitrosamines in mainstream cigarette smoke by gas chromatography/ion trap mass spectrometry.
A specific and sensitive method was developed and validated to quantitatively analyze four tobacco-specific nitrosamines (TSNAs) in the particulate phase of mainstream cigarette smoke. Cigarette smoke particulate matter was collected according to ISO 4387. The particulate matter was extracted with acetic ether, cleaned up with a Supelclean ENVI-Carb silod-phase extraction (SPE) cartridge, concentrated under the protection of nitrogen and analyzed by gas chromatography (GC)/ion trap mass spectrometry (MS) with a very-low-flow-loss column (VF-17 MS) in MS(n) mode using N-nitrosopentyl-3-picolylamine (NNPA) as an internal standard. ⋯ The method achieved excellent reproducibility (RSD: 1.8-4.8% for intra-assay, 3.4-7.1% for inter-assay). It also shows no evidence of artifact formation. This method is very suitable for the routine detection of TSNAs in mainstream cigarette smoke.

Simultaneous determination of docetaxel and ketoconazole in rat plasma by liquid chromatography/electrospray ionization tandem mass spectrometry.
A rapid and sensitive liquid chromatography/tandem mass spectrometry (LC/MS/MS) method has been developed and validated for simultaneous quantification of docetaxel and ketoconazole in rat plasma with paclitaxel as internal standard (IS). The analytes were extracted from rat plasma by using a liquid-liquid extraction technique with ethyl acetate and the LC separation was performed on a Cosmosil-C(18) analytical column (150 mm x 2.0 mm i.d., Nacalai Tesque Inc., Japan). The extracted samples were analyzed with LC/MS/MS, operating in selected reaction monitoring (SRM) mode. ⋯ The total analysis time was only 9.0 min. This quantitation method was successfully applied to the simultaneous determination of docetaxel and ketoconazole in rat plasma and some potential interaction was found in the current coadministration pharmacokinetic study. This established method was also utilized in the in vitro and in vivo drug-drug interaction study of docetaxel and ketoconazole (to be published).
